基于癌症基因组图谱构建膀胱癌免疫基因预后评估模型

Establishing a prognostic model for bladder cancer by immune genes based on cancer genome map

  • 摘要: 目的 采用生物信息学方法,构建膀胱癌患者免疫基因预后评估模型,筛选影响膀胱癌发生及预后的生物标志物。方法 下载癌症基因组图谱(The Cancer Genome Atlas,TCGA)数据库中膀胱癌及癌旁正常组织转录组数据及临床病理资料,下载免疫信息学数据库(Immunomes Project,ImmPort)中免疫基因数据,筛选差异表达免疫基因;采用单因素、多因素Cox风险回归模型分析并构建差异表达免疫基因预后评估模型。进一步分析免疫基因预后评估模型风险评分、临床病理特征与膀胱癌预后的相关性。结果 基于R语言中"wilcox.test"共筛选出260个与膀胱肿瘤相关的差异表达免疫基因;纳入Cox单因素分析,结果显示25个差异表达免疫基因与膀胱肿瘤预后相关(P<0.01),取P<0.01的25个差异表达免疫基因纳入Cox多因素分析,得到包含11个(TAP1、MMP9、ADIPOQ、PDGFRA、AHNAK、KCNH2、OLR1、RAC3、NAMPT、PPY、SH3BP2)差异表达免疫基因的膀胱癌免疫基因预后评估模型;基于模型风险评分将样本分为高、低风险组,Kaplan-Meier生存曲线结果显示高风险组5年总生存率为28.6%,低风险组5年总生存率为59.7%。ROC模型曲线证实该预测模型有一定的准确性(AUC=0.732)。将临床病理特征纳入Cox回归分析,结果显示免疫基因预后评估模型评分为高风险是影响膀胱癌患者预后的独立预测因子。结论 基于生物信息学方法构建的膀胱癌免疫基因预后评估模型可用于计算膀胱癌患者预后风险评分,期望为临床判断膀胱癌患者的预后提供帮助。

     

    Abstract: Objective To establish a bioinformatics model for evaluating the prognosis of bladder cancer by immune genes,and screen the biomarkers influencing the occurrence and prognosis of bladder cancer. Methods The transcriptome data and clinicopathological data of bladder cancer and adjacent normal tissues were downloaded from the Cancer Genome Atlas database,immune gene data were downloaded in the ImmPort database,and the differentially expressed immunogenes were screened.Univariate and multivariate Cox regression models were used to analyze the prognosis of bladder cancer by differentially expressed immune genes.The correlation between risk score,clinical and pathological features of bladder cancer and prognosis of bladder cancer were further analyzed. Results Totally 260 differentially expressed bladder tumor-related immune genes were screened out based on "Wilcox.Test" in R language.Univariate Cox analysis showed that 25 differentially expressed immune genes were related to the prognosis of bladder tumor (P<0.01).Then the genes were inputted in the multivariate Cox regression analysis,and a prognosis model including 11 immune genes (TAP1,MMP9,ADIPOQ,PDGFRA,AHNAK,KCNH2,OLR1,RAC3,NAMPT,PPY,SH3BP2) differentially expressed in bladder cancer was achieved.The samples were divided into high risk group and low risk group by the median value of the model risk score.Kaplan-meier survival curve results showed that the 5-year overall survival rate (OS) of the high risk group was 28.6%,and that of the low risk group was 59.7%.The ROC curve analysis confirmed the accuracy of the prediction model (AUC=0.732).The clinicopathological features were included in Cox regression analysis,and the results showed that the high risk score of the immune gene prognostic assessment model was an independent predictor of the prognosis of patients with bladder cancer. Conclusion The bladder cancer prognosis model based on immune genes is reliable,which is expected to help clinical physicians to predict the prognosis of bladder cancer.
