宫颈癌及癌前病变中Aurora-A的表达特征及与HR-HPV感染相关性

Expression of Aurora-A in cervical cancer and precancerous lesions and its correlation with HR-HPV infection

  • 摘要: 目的 分析各类宫颈组织中Aurora-A表达特征及与高危型人乳头瘤病毒(high risk-human papilloma virus,HR-HPV)感染的相关性。方法 选取2016年3月-2019年8月存于深圳市第二人民医院(深圳大学第一附属医院)的正常宫颈组织22份、低级别鳞状上皮内病变(low squamous intraepithelial lesion,LSIL)组织22份、高级别鳞状上皮内病变(high squamous intraepithelial lesion,HSIL)组织44份、宫颈癌(cervical cancer,CC)组织22份,免疫组织化学检测Aurora-A蛋白在各类宫颈病变组织的表达,分析Aurora-A蛋白及HR-HPV感染与病变程度的相关性。结果 与正常宫颈组织相比,Aurora-A在LSIL、HSIL及CC组织中过表达(P均<0.05),且其阳性率随着宫颈癌变程度的升高而升高,呈正相关(r=0.486,P=0.032);HR-HPV感染率随着宫颈癌变程度的升高而升高,呈正相关(r=0.596,P=0.029);Aurora-A表达与HR-HPV感染率呈正相关(r=0.625,P=0.023)。结论 Aurora-A过表达可能与宫颈癌发生相关,或可作为诊断鳞状上皮内病变及CC的参考指标。

     

    Abstract: Objective To analyze the expression of Aurora-A in cervical cancer and precancerous lesions and its correlation with high risk-human papilloma virus (HR-HPV) infection. Methods A total of 110 cervical specimens were collected from the Shenzhen Second People's Hospital (the First Affiliated Hospital of Shenzhen University),with 22 cases had normal cervical tissues,22 cases had low-grade squamous intraepithelial lesions (LSIL),44 cases had high-grade squamous intraepithelial lesions (HSIL) and 22 cases had cervical cancer (CC).Aurora-A protein expression in various cervical specimens were detected by immunohistochemistry assay.The correlation of Aurora-A protein and HR-HPV with the degree of cervical lesions was analyzed. Results Compared to normal cervical tissue,Aurora-A was over-expressed in LSIL,HSIL and CC tissues (all P<0.05),and its positive rate increased with the degree of cervical lesions (r=0.486,P=0.032).There was also a positive correlation between the HR-HPV and the degree of cervical lesions (r=0.596,P=0.029).Aurora-A and HR-HPV were positively correlated (r=0.625,P=0.023). Conclusion Aurora-A overexpression may be related to the occurrence of cervical cancer,and may be used as a reference index for the diagnosis of squamous intraepithelial lesion and CC.
