脑肌酸激酶/磷酸肌酸系统与中枢认知功能的机制综述

Review on mechanisms of brain creatine kinase/phosphate creatine system and central nervous cognitive function

  • 摘要: 肌酸激酶(creatine kinase,CK)是脑能量代谢过程中的重要催化酶之一,在磷酸肌酸和三磷酸腺苷的高能磷酸基团转移过程中发挥着重要作用。CK与磷酸肌酸构成的能量缓冲系统具有良好的神经保护作用,该系统的异常与认知功能障碍密切相关,而肌酸增补或激活CK对认知功能障碍有一定的改善作用。本文对近年来相关的研究进行综述,以阐明认知功能障碍的病理生理机制,并为以肌酸或肌酸激酶为靶点开发新药提供一定的参考。

     

    Abstract: Creatine kinase is one of the most important catalytic enzymes in brain energy metabolism and plays an important role in transferring high-energy phosphates between creatine phosphate and adenosine triphosphate. The energy buffer system consisting of creatine kinase and phosphocreatine has good neuroprotective effect. The abnormality of this system is closely related to cognitive dysfunctions, while creatine supplementation or creatine kinase activation may improve cognitive dysfunction. This article summarizes related researches during these years, so as to further clarify the pathophysiological mechanism of cognitive dysfunctions and provide a feasible reference for the development of new drugs targeting creatine or creatine kinase.
