Abstract:
Objective To study the effect of human telomerase reverse transcriptase(hTERT) gene on proliferation of in vitro cultured human hepatocytes and expression of cytochrome P450 19A1.
Methods Recombinant retrovirus vector carrying pLNCX2-hTERT fragment was transfected into in vitro cultured human hepatocyte.Effect of hTERT gene on proliferation of human hepatocytes and expression of P450 19A1 was detected by methyl thiazol tetrazolium(MTT) and Western blot,respectively.
Results The retrovirus vector carrying pLNCX2-hTERT fragment was successfully transfected into human hepatocytes.The proliferation of human hepatocytes was significantly faster after transfection than before transfection.Hepatocytes at 55kU expressed CYP450 19A1 protein before and after transfection.
Conclusion Exogenous hTERT gene can promote the proliferation of human hepatocytes,both transfected or untransfected hepatocytes can express CYP450 19A1.
