Abstract:
Objective To study the effect of temperature–responsive chitosan hydrogel on biological performance of adipose-derived stem cells(ADSC).
Methods After ADSC were isolated and temperature-responsive chitosan hydrogel was prepared, biological performances of ADSC,including their vitality, proliferation and multipotential differentiation, were detected in temperature–responsive chitosan hydrogel in vitro.
Results ADSCs were successfully cultured and confirmed by flow cytometry. When cocultured with chitosan hydrogel,ADSCs could grow very well and retain the biological performance of stem cells. After directional induction, they were able to further differentiate into cardiomyocyte-like cells,as well as adipogenic and osteogenic lineages.
Conclusion Temperature–responsive chitosan hydrogel has a good biocompatibility, and can thus be used as an injectable bypass material in cardiac muscle engineering.
