大鼠腹主动脉狭窄高血压心肌肥厚模型的优化

Improvement of cardiac hypertrophy model induced by abdominal aortic coarctation in rats

  • 摘要: 目的: 优化大鼠腹主动脉狭窄高血压心肌肥厚模型的复制方法。方法: 采用腹部左侧纵切口手术方法制备腹主动脉狭窄高血压心肌肥厚大鼠模型,动态监测大鼠尾动脉压,术后4周检测颈总动脉压、左心室内压,称重法测定心脏系数和心肌肥大系数,并进行组织学观察。结果: 与假手术组比较,腹主动脉狭窄手术组大鼠2周后尾动脉压升高29%,4周时颈动脉压和心室内压分别增加28%和36%(均为P<0.01),心脏指数和心肌肥大指数分别增大0.86和0.89(均为P<0.01)。病理切片显示心肌细胞肥大。结论: 腹部左侧纵切口术式制备大鼠腹主动脉狭窄高血压心肌肥厚模型,方法简便可靠,重复性好,成功率高

     

    Abstract: Objective: To optimize the cardiac hypertrophy model induced by abdominal aortic coarctation in rats. Methods: Using left side direct entering approach, the hypertensive hypertrophy was induced with abdominal aortic coarctation in rats. The tail artery blood pressure was measured dynamically before and after operation. At the end of the experiment of 4 weeks,carotid and left ventricular pressures,cardiac and hypertrophy coefficient were measured. The hearts were observed histologically. Results: Compared with sham-operated animals,the tail artery blood pressure in operated rats increased by 29%(P <0.01) two weeks after operation. By four weeks,carotid blood pressure and left ventricular pressure of the operated group increased 28% (P <0.01)and 36% (P <0.01),respectively. Cardiac coefficient (heart weight/body weight) and cardiac hypertrophy coefficient(left ventricle weight/heart weight) increased 0.86 (P <0.01) and 0.89 (P <0.01) respectively,compared with sham-operated rats. The histological slices showed hypertrophied cardiac myocytes. Conclusion: The left side direct entering approach can reliably duplicate hypertensive hypertrophy model in rats with high repeatability and success rate.
