呼吸道鼻病毒、SARS病毒的Teflo滤膜采集及RT-PCR检测的初步研究

  • 摘要: 目的: 建立Teflo滤膜采集检测呼吸道鼻病毒及SARS病毒颗粒的技术模型,初步检测分析我院重点病区环境(病房内)空气的SARS病毒是否存在及分布情况。方法:利用Teflo滤膜过滤式采集病房空气中的鼻病毒及SARS病毒,然后采用逆转录PCR方法,从滤膜分离标本中进行病毒特异性基因的扩增(鼻病毒5′端非编码区;SARS病毒聚合酶基因)。结果:准确检测到重症哮喘患者房间内的鼻病毒;我院重点病区环境包括病房内空气中未检测到SARS病毒的存在.结论: Teflo滤膜/RT-PCR方法用于医院病区空气中SARS病毒及其他呼吸道病毒颗粒的监测可行性强,我院重点病区无SARS病毒分布。

     

    Abstract: Objective:to build up approach of sampling the respiratory tract viruses in the air with Teflo filter and RT-PCR analysis,further monitor the possible existence of SARS virus in the key clinical sections of our hospital.Methods:The air of different sampling points was filtrated with Teflo filter and the rhinovirus or possible SARS virus particles were collected.The viral RNA was extracted and the sem-i nested RT-PCR for 5′-uncoding gene region of rhinovirus or one-step RT-PCR for polymerase gene of SARS virus was performed,respectively.Results:The filter samples in the ward of patient with severe asthma syndrome were rhinovirus-positive;in contrast,the filter samples of all three clinical sections of our hospital were SARS virusnegative. Conclusions:The Teflo-filter air sampling and extraction methodology developed in this study should be applicable to the detection of rhinovirus,SRAS virus and other airborne human respiratory tract viruses in the indoor air of hospital wards.
