应用激光扫描共聚焦显微技术观察PDT导致癌细胞内钙水平的变化

Changes of intracellular calcium level in GLC 82 cells induced by PDT: laser scanning confocal study

  • 摘要: 目的: 肿瘤光动力治疗(photodynamic therapy,PDT)是现代癌症治疗中一种新的治疗法,为了更好地认识光动力诱导肿瘤细胞凋亡的作用机制。方法: 用Fluo-3AM荧光探针负载培养的人肺腺癌GLC-82细胞,在激光扫描共聚焦显微镜下观察细胞内Ca2+浓度的变化。结果: 激光单独照射前后GLC-82细胞内Ca2+的浓度变化不大;在PDT作用下,激光照射前GLC 82细胞内Ca2+的浓度变化明显。结论: 本方法灵敏、简单、快速,能实时监测细胞内游离钙的变化。

     

    Abstract: Objective: Photodynamic therapy (PDT) has become an important new modality in cancer treatment. In order to obtain a better understanding of the mechanisms of apoptpsis induced by PDT. Methods: Cultured GLC-82 cells were loaded with fluo-3/AM fluorescence probes, and the changes of intramyocytic Ca2+ concentration were examined by laser scanning confocal microscopy. Results: there were negligible changes of intracytoplasmic calcium concentration of GLC-82 cells after laser irradiation alone. There were remarkble changes of intracytoplasmic calcium concentration of GLC-82 cells after laser irradiation. Conclusion: it is sensitive, rapid, simple and most effective method for real time quantitation of intracellular free calcium.
