三氧化二砷对Hela细胞hTERTmRNA表达的调节和对端粒酶活性的影响

The effect of As2O3 on telomerase activity and human telomerase reverse transcriptase mRNA in Hela cells

  • 摘要: 目的: 观察三氧化二砷 (As2O3)对人宫颈癌细胞 (Hela细胞)端粒酶活性及端粒酶逆转录酶 (hTERTmR NA)表达的影响。方法: 以 2 μmol/L的As2O3 作用于人宫颈癌Hela细胞, 在不同时间点用MTT法检测Hela细胞活力的变化, 用流式细胞仪检测细胞周期的变化, 用RT PCR法检测hTERTmRNA表达的变化, 用TRAP ELISA法检测端粒酶活性的变化。结果: 随着As2O3 作用时间的延长, Hela细胞的活力逐渐降低, G0/G1期细胞所占比例升高, S期细胞所占比例减少, hTERTmRNA表达水平逐渐降低, 端粒酶活性逐渐下降。结论: As2O3 可引起Hela细胞周期阻滞, hTERTmRNA的表达水平下调, 端粒酶活性降低。

     

    Abstract: Objective:To investigate the impact of Arsenic Trioxide on telomerase activity and human telomerase reverse transcriptase (hTERT) in HeLa cell. Methods: The Hela cells were cultured and exposed to 2μmol/L As2O3 during different time, at the different time point, the cell's proliferation activity were measured by MTT, the distribution of cell cycle were determined by flow cytometry, the expression of hTERT mRNA were measured by RT-PCR, the activity of telomerase were measured by TRAP-ELISA. Results: The Hela cell's proliferation activity was gradually decreased after exposed to 2μmol/L As2O3, the number of cells in the GO/G1 phase increased and S phase decreased. The expression of hTERT mRNA was gradually weakened and the telomerase activity was decreased. Conclusions: As2O3 has cells cycles stopping effection on HeLa cell, and it can weaken the expression of human telomerase reverse transcriptase (hTERT) and decreased the telomerase activity.
