低温储存对皮肤组织E-钙黏素和β-连接素表达的影响

Expression of E-cadherin and β-catenin in cryopreserved human skins

  • 摘要: 目的: 观察黏附分子E-钙黏素(E-cadherin)、β-连接素(β-catenin)在不同温度储存皮肤组织中的分布,以探讨其与皮肤冷冻损伤的关系。方法: 用E-cadherin、β-catenin抗体对5种储存条件下的皮肤作免疫组化染色,用图像分析仪测定表皮组织中的含量。结果: 与新鲜皮片组相比,-196℃ H抗冻液中储存皮肤组织结构保存较好,E-cadherin、β-catenin含量变化不明显,而4℃、-20℃、-80℃储存皮肤组织结构破坏明显,含量亦明显下降。结论: 皮肤低温储存冷冻损伤与黏附分子破坏有关。

     

    Abstract: Objective: To find out the distribution of E-cadherin and β-catenin in cryopreserved skins and investigate the relationship with cryoinjury. Methods: Immunohistochemical staining with antibodies of E-cadherin and β-catenin in 5 groups of skins. Results: Comparing to the normal human skin, The histological structure and E-cadherin、β-catenin level of skins stored at -196℃ in H anti-frozen liquid did not change significantly, while those in groups kept at 4℃、-20℃、-80℃ were destroyed. Conclusion: The skin cryoinjury is correlated with cell adhesion molecules.
