摘要: 目的: 探讨支气管上皮细胞活化对ICAM-1基因和细胞表面ICAM-1蛋白质表达的影响。方法: 用10ng/m l TNF-α作用于正常培养的支气管上皮细胞(BEAS-2B)12h,通过RT-PCR分析ICAM-1在BEAS-2B细胞中的基因表达和用流式细胞仪分析ICAM-1在BEAS-2B细胞表面蛋白质量。结果: TNF-α与BEAS-2B细胞共同培养12h,可上调BEAS-2B细胞ICAM-1基因的表达和增加BEAS-2B细胞表面的ICAM-1蛋白质量。结论: TNF-α可诱导BEAS-2B细胞ICAM-1基因和蛋白质表达。Abstract: Objective:To study the effect of epithelial cell activation on the gene and cell surface expression of ICAM-1.Methods:BEAS-2B cells were activated by TNF-α(10 ng/ml) for 12h,ICAM-1 gene in BEAS-2B cells were analyzed by RT-PCR and cell surface ICAM-1 protein was assayed by Flow cytometry.Results:TNF-α upregulated ICAM-1 gene expression in BEAS-2B cells and increased ICAM-1 protein on BEAS-2B cell surface.Conclusion:TNF-α might induce BEAS-2B cells to express ICAM-1.
