结核杆菌embB基因突变与临床治疗的关系

The relationship of the mutation on embB genes and drug-resistence with clinical treatment

  • 摘要: 目的: 了解结核菌耐乙胺丁醇药敏实验和embB基因突变情况,研究其临床应用价值。方法: 通过聚合酶链反应(PCR)-单链构象多态性(SSCP)技术初步鉴定96株分支杆菌临床分离株的菌种,并进一步分析其embB基因。结果: 分析56株分支杆菌临床分离株的16SrDNASSCP电泳图谱均与结核分支杆菌标准株相同。21株药物敏感株的embB基因SSCP均泳动正常;35株耐乙胺丁醇分离株中,17株(4 8.6%) embB基因SSCP泳动异常。结论: 部分结核分支杆菌耐乙胺丁醇是由于其embB基因突变所致。且embB基因突变均发生在药敏实验高浓度区。

     

    Abstract: Objective: To realize the relevance of the mutations of embB genes with drug resistence in M. tuberculosis isolates,and to evaluate their clinical value. Methods: 96 clinical isolates were identified their mycobacterial species,and then analyzed their embB genes with PCR SSCP. Results: 56 clinical isolates all had the same SSCP profiles as M.tuberculosis. 21 drug-sensitive isolates had normal embB SSCP profiles. Of 35 ethambutol-resistant isolates,17(48.6%) displayed abnormal embB SSCP profiles. Conclusions: Ethabutol resistancesin some M.tuberculosis isolates were due to mutations on embB genes,Moreover,the mutation of embB genes has high accidence in isolates which resistanthigh concentration ethambutol.
