脑微血管内皮细胞、星形胶质细胞共同培养建立血脑屏障模型

To establish in vitro blood-brain barrier model by coculturing brain microvessel endothelial cells and astrocytes

  • 摘要: 目的: 建立能够模拟在体血脑屏障的体外模型。方法: 培养大鼠脑微血管内皮细胞和星形胶质细胞,用免疫组织化学染色的方法鉴定细胞,分别接种两种细胞于多聚酯多孔膜的两面,相差显微镜及HE染色观察细胞在多聚酯膜上的生长情况。结果: 培养的脑微血管内皮细胞多数为梭形铺路石样外观,而星形胶质细胞呈具有多个突起的典型形态,免疫组织化学染色鉴定细胞的纯度分别在90%和95%以上,两种细胞能够以单层细胞的形式生长于多聚酯多孔膜的两面。结论: 通过脑微血管内皮细胞和星形胶质细胞共同培养能够形成与在体血脑屏障类似的结构,是一种建立血脑屏障模型的可行方法。

     

    Abstract: Objective: To establish in vitro blood-brain barrier model. Methods: Rat brain microvessel endothelial cells and astrocytes were cultured and immunohistochemical stainning were employed to identify the cells. The two kinds of cell were then seeded onto each surface of polyester-multipore-membrane. Growth condition of the two kinds of cell on the membrane was evaluated by phase contrast microscope and HE stainning. Results: The two kinds of cultured cellpresent typical shape of brain microvessel endothelial cell and astrocyte respectively.Immunohistochemical stainning proved that the purity of the two kinds of cell was above 90% and 95%. The two kinds of cell both formed continued monolayer on each surface of the polyester-multipore-membrane. Conclusions: Coculturingbrain microvessel endothelial cells and astrocytes on polyester-multipore-membrane resemle in vivo blood-brain barrier closely, which can be used as in vitro blood-brain barrier model.
