Abstract:
Objective To study the effect of oxidized low density lipoprotein(LDL)on placental growth factor-1(PGF-1)influencing releasing of inflammatory cytokines in human umbilical vein endothelial cells(ECV-304).
Methods Under the condition of oxidized LDL,ECV-304 cells(2×105)were incubated in RPMI-1640 medium containing 2%fetal bovine serum in the absence or presence of PGF-1(20,40,and 80ng/mL)for 3,6,12 and 24h,respectively.IL-6 and MMP-2 levels in the supernatant were measured using an ELISA quality kit.
Results PGF-1 induced the secretion of inflammatory cytokines in a dose-and time-dependent manner.The expression level of IL-6 and MMP-2 was higher in ECV-304 cells under the condition than not under the condition of oxidized LDL.
Conclusion Oxidizing injury is an independent risk factor for artherosclerosis.PGF can up-regulate the expression of inflammatory cytokines,and further deteriorate the disease.
