大鼠长期应用复方樟柳碱的病理学观察

The pathological study on rats treated with compound anisodine

  • 摘要: 目的:观察大鼠长期应用复方樟柳碱的中毒反应及可逆性,寻找中毒的靶器官。方法:选用Wistar大鼠120只,随机分4组,每组30只。分别设为生理盐水对照组、复方樟柳碱每天8.08mg/kg、40.4和323.2mg/kg组。在连续皮下注射给药6周后,各组分别处死2/3的动物,余下动物停药并继续观察2周后处死,实验过程中做以下检查:一般生理观察、心血细菌培养、8~9项脏器重量系数及对24个脏器(对照组、40.4及323.2mg/kg组所有动物)的病理组织学检查。结果:8.08及40.4mg/kg组动物,除后一组给药部位皮肤出现轻微结痂外,与对照组相比无明显差异。323.2mg/kg组明显中毒,主要表现为给药局部组织坏死、溃疡或结痂,动物精神沉郁、体重增长迟缓及部分动物(7~8只)给药后出现短暂的抽搐反应。此外,由于动物体表组织大面积坏死、溃疡,导致该组动物近半数出现菌血症,少数动物皮下感染向深层发展并波及腹腔脏器,有4只动物在实验过程中死亡。结论:复方樟柳碱中毒的靶器官为给药局部组织和中枢神经系统。

     

    Abstract: Objective:To observe the toxic reactions and the toxic target organs of rats treated with compound anisodine.Methods:The toxic pathological changes were evaluated in Wistar rats subcutaneously administrated with compound anisodine 8.08, 40.4 and 323.2 mg/kg daily for 6 weeks. Results:Except for the slight scab apeared at the injected site in the 40.4 mg/kg group, there were no other toxic effects in the 8.08 and 40.4 mg/kg groups. However, in the 323.2 mg/kg group, the toxic responses including local necrosis, ulceration and scar formation at the administrated area of the skin occurred. The animals were inactive and body weight increased slowly. About one-fourth (8/30) rats of this group showed clinic convulsion following injection. In addition, bacteremia revealed in nearly half of the animals because of the large area of skin necrosis and ulceration. The cutaneous infection also spread to the abdominal organs.Conclusion:This study suggests that the injection area and the central nerves systerm maybe the toxic target organs of compound anisodine.
