血管紧张素转换酶抑制药对慢性缺氧大鼠血循环内皮细胞数量的影响

Effect of angiotensin-converting enzyme inhibitor on circulating endothelial cells in rats with chronic hypoxic pulmonary hypertension

  • 摘要: 目的 本研究旨在观察血管紧张素转化酶抑制剂(ACEI)西拉普利对慢性缺氧性肺动脉高压大鼠循环内皮细胞(CEC)数量及形态的影响,探讨对血管内皮的保护作用机制。方法 观察大鼠慢性缺氧性肺动脉高压的血液动力学变化及循环内皮细胞、血管紧张素转化酶、脂质过氧化物的变化。结果 缺氧性肺动脉高压时肺动脉压力升高、脂质过氧化物产生丙二醛(LPO)升高,血管紧张素转化酶(ACE)活性降低,循环内皮细胞数量增加(P<0.01);西拉普利治疗后脂质过氧化物降低,血循环内皮细胞数量明显下降(P<0.01)。结论 本实验表明ACEI可以抑制氧自由基反应及脂质过氧化、平衡ACE系统从而减少血CEC数量、降低肺动脉压而达到保护血管内皮功能。

     

    Abstract: Objective To investigate the protective effects of angiotensin-converting enzyme(ACE) inhibitor(cilazapril) on circulating endothelial cells(CEC) in rats with chronic hypoxic pulmonary hypertension. Methods Catheter techniques were used to observe the hemodynamic change,microscopy was performed to observe the changes of CEC,and biochemical methods were employed to determine lipid peroxidization(LPO) and ACE. Results The levels of LPO,CEC,right ventricular pressure,systolic pulmonary arterial pressure,and mean pulmonary arterial pressure were increased in the hypoxia group and decreased in the ACE inhibitor(cilazapril) group compared with the placebo group(P<0.01). Conclusion Cilazapril as an ACE inhibitor can reduce CEC by inhibiting LPO.
