血管内放射抑制猪冠状动脉球囊损伤后血管重塑的机制

Endovascular radiation inhibits neointima formation after coronary artery balloon injury in swine

  • 摘要: 目的: 探讨血管内放射对冠状动脉内介入治疗后血管重塑的抑制作用。方法: 通过小型猪冠状动脉球囊扩张术后再狭窄动物模型,采用形态学、组织学方法和α平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)免疫组织化学染色方法,观察了192 Ir血管内放射治疗对血管重塑的影响。结果: 血管内放射治疗后分别显著增加了球囊扩张术后30d血管周长、血管腔周长和管腔面积,血管内放射显著减少了术后30d外膜α-SMA阳性细胞及外膜纤维化。结论: 血管内放射治疗可能通过通过减少外膜的α-SMA阳性细胞及血管外膜纤维化,抑制球囊扩张术后血管重塑的过程。

     

    Abstract: Objective: To determine if Endovascular radiation will be effective in inhibiting vascular remodeling after coronary intervention. Methods: Twelve swine underwent coronary balloon overstretch. The radiation delivery system was used to deliver 20 Gy to dilated segment in the seven swine. All animals were given aspirin (325 mg) daily and were killed at 30 days. Cross sections were used to perform computer-based morphometric analysis and immunohistochemistry(α-actin) stain. Results: Vessel perimeter, lumen perimeter, and lumen area were increased significantly in the irradiated segments. There were fewer α-actin positive cells in the adventitia of the irradiated vessels than in nonirradiated controls. Conclusion: Endovascular radiation demonstrated a favorable effect on late remodeling by reducing α-actin positive cells and preventing the fibrosis in the adventitia.
