Abstract:
Objective To observe the effect of RhoB on NF-κB transcriptional activity in human pancreatic cancer SW1990 cells.
Methods Transcription activity of genes was detected by double-luciferin and RhoB protein was detected by Western blot 24 h after zero-load plasmid(pcDNA3),wild-type RhoB over-expression plasmid(RhoB-wt),RNA interference control plasmid(RNAi-control),RhoB RNA interference plasmid(RhoB-RNAi),RhoB-activated plasmid(RhoB-V14) or RhoB-inactivated plasmid(RhoB-N19),NF-κB reporter gene plasmid(NF-κB-luc),and intra-reference plasmid(pRL SV40-luc) were transfected into SW1990 cells.
Results The over-expression and activation of transfected RhoB(0.8μg) could inhibit the NF-κB reporter gene activity in SW1990 cells by 54% and 21% compared to the control group.The transcriptional activity of NF-κB was 1.5-fold higher in SW1990 cells transfected with RhoB than in the control group.
Conclusion RhoB can inhibit the transcriptional activity of NF-κB in pancreatic cancer SW1990 cells.
