小型无绿藻的形态与显微结构观察

Micro structural investigation on Prototheca wickerhamii

  • 摘要: 目的:对从1名女患者面部皮肤结节活检分离出的1株无绿藻菌,从形态学的角度上对该株菌进行较全面的观察。方法:通过分离培养鉴定、直接涂片镜检,扫描电镜、透射电镜对其超微结构进行观察,组织病理切片观察。结果:菌体透明,细胞壁厚,并见到内有1~10内生孢子的孢子囊。HE染色在真皮浅层病灶区可见许多形如“桑葚状”的大孢子,壁厚,直径3~11μm,内见球形或卵圆形内生孢子。结论:从形态学特征上确定病原菌是一株小型无绿藻。

     

    Abstract: Objective: To observed the morphological characteristics of a strain of Prototheca isolated from a nodule specimen on the face of a woman patient.Methods: The strain of Prototheca was directly smeared, isolated and cultured, examined under optic, scanning, and transmitting electronic microscopes for its ultra-structural characterestics. The tissue sections were studied histophathogenically for its pathogenicity.Results: The P.wickerhamil was demonstrated as hyaline, globular or ovoid cell, in which a number of round, ovoid unicellular spores ranging from 1 to 10 could be seen. Numerous morula, daisy-like sporangia could be seen in the superficial layer of dermis in HE stained sections, which were thick-walled, ranged in diameter from 3 to 11 μm, with ovoid or sphenical endospores inside.Conclusion: Based on the microbiological and histopathological examinations, the pathogen of this case was confirmed as P. wickerhamii.
