亚低温预处理大鼠局灶性脑缺血的保护作用

Experimental study on the preconditioning therapy for protection against focal ischemia in rats by mild hypothermia

  • 摘要: 目的:模拟神经外科手术缺血过程,研究亚低温预处理情况下大鼠局灶性脑缺血的耐受时间。方法:采用线栓法制作可逆的大鼠大脑中动脉缺血再灌注模型。72只SD大鼠随机分成6组:1h、2h、3h缺血-常温组,1h、2h、3h缺血-亚低温预处理组。术后评价大鼠的神经功能以及脑梗塞体积的变化。结果:对于1h和2 h缺血组,应用亚低温预处理,可以改善大鼠的神经功能恢复、减小脑梗塞体积。对于3h缺血组,则脑保护作用不明显。结论:应用亚低温预处理大鼠局灶性脑缺血,可以减轻脑缺血性损害,脑缺血的耐受时间应在2h以内。

     

    Abstract: Objective: To simulate the ischemia of neurosurgery and study the duration of brain ischemia in rats by preconditioning therapy of mild hypothermia.Methods: Reversible middle cerebral artery occlusion (MCAO) was induced by employing a intraluminal suture.SD rats were assigned randomly to one of six groups (n=12) as follows: 1hr,2hrs,3hrs MCAO-normothermic group and 1h,2h,3h MCAO-mild hypothermia group.Neurological examinations,infarcted volume.Results: In 1 h and 2h MCAO groups,applying mild hypothermia before the ischemia can improve the neurological outcome and reduced the infarcted volume.In 3h MCAO-mild hypothermia groups,there is no effect of brain protection.Conclusion: Preconditioning therapy of mild hypothermia can reduce the injury of brain ischamia,the duration of brain ischemia is with in 2hrs.
