阿尔茨海默病患者体内端粒长度与氧化应激关系

Correlation between oxidation stress and telomere length in patients with Alzheimer’s disease

  • 摘要: 目的 探讨阿尔茨海默病(Alzheimers'disease,AD)患者与脂质过氧化应激相关的端粒长度变化。方法 40名AD患者,59名年龄相当的正常对照。分别测定所有受试者尿液中8-异前列腺素F2α(8-iso-prostaglandin F2alpha,8-iso-PGF2α)的浓度,外周血单核细胞端粒长度,包括端粒平均长度与端粒长度分布。结果 AD患者尿8-iso-PGF2α的水平明显高于对照组,(71.68±2.60)ng/mmol vs(46.90±1.88)ng/mmol creatinine(P<0.001),端粒平均长度差异无统计学意义,但稍长端粒(4.4-9.4kb)比例明显增加,短端粒(<4.4kb)比例减少。结论 AD患者体内氧化应激较对照组增高;端粒长度分布呈现氧化应激相关的变化,最短端粒对氧化应激更敏感。

     

    Abstract: Objective To study the lipid peroxidation-related changes of telomere length in patients with Alzheimer’s disease(AD).Methods Forty AD patients(experimental group) and 59 healthy subjects(control group) were enrolled in this study.Concentration of 8-iso-prostaglandin F2α(8-iso-PGF2α) in urine and telomere length in peripheral mononuclear cells(including average length of telomere and its distribution) were measured,respectively.Results The urinary 8-iso-PGF2α level was statistically higher in AD patients than in controls(71.68±2.60)ng/mmol vs(46.90±1.88)ng/mmol creatinine(P<0.001) with no difference observed in the mean length of telomeres.However,the ratio of slightly longer telomeres(4.4-9.4kb) was increased,while that of short telomeres(<4.4kb) was decreased in AD patients.Conclusion The oxidation stress level is statistically higher in AD patients than in controls.The distribution of telomere length is related with the oxidation stress-related change.The shortest telomeres are more sensitive to oxidation stress.
