实验性肝纤维化形成和逆转过程中基质金属蛋白酶-13蛋白及其mRNA表达

Expression of matrix metalloproteinase13 and its mRNA in carbon-tetrachoride-injured liver fibrosis and its reversal

  • 摘要: 目的: 研究基质金属蛋白酶13(matrix metalloproteinase 13,MMP-13)在大鼠四氯化碳实验性肝纤维化形成和逆转过程中的表达。方法: 应用免疫组织化学和核酸原位杂交方法研究MMP-13蛋白及其mRNA在四氯化碳实验性大鼠肝纤维化形成和逆转过程中的表达变化,并进行网织纤维染色和胶原纤维染色。结果: MMP-13蛋白及其mRNA在肝脏间质细胞和部分肝细胞中表达。在纤维化形成过程中MMP-13蛋白及其mRNA的表达量相对增加;在纤维化逆转过程中MMP-13蛋白及其mRNA的表达量是由强渐弱的动态变化。结论: 肝脏间质细胞是MMP-13蛋白及其mRNA的主要细胞来源,MMP-13在肝纤维化形成和逆转过程中具有重要作用。

     

    Abstract: Objective: To explore the expression of matrix metalloproteinase 13(MMP-13) and its mRNA in carbon-tetrachoride-injured liver fibrosis and its reversal. Methods: By immunohistochemical analysis and nucleic acid in situ hybridization(ISH) we observed the expression of MMP-13and its mRNA in specimens from liver of Wistar rats induced by carbon tetrachloride. Results: Expression of MMP 13and its mRNA was found mainly in mesenchymal cells,also in parts of hepatocytes.The levels of MMP-13expression were increased in the early time of liver fibrosis and decreased gradualy in the reversal of liver fibrosis. Conclusions:Mesenchymal cell is the main cellular origin of MMP13and its mRNA. MMP 13may play an important role in the liver fibrogenesis and its reversal.
