蛋清溶菌酶对人单纯疱疹病毒Ⅰ型抑制作用的实验研究

Study of inhibitive effect of hen egg-white lysozyme on herpes simplex virus type 1

  • 摘要: 目的:观察蛋清溶菌酶(HEWL)对人单纯疱疹病毒I型(hHSV-1)的抑制作用。方法:以HSV-1的敏感细胞BHK-21为实验细胞模型,以更昔洛韦为阳性对照药,以0.5%12-烷基磺酸钠(SDS)为阻断剂,采用细胞病变法(CPE)判定溶菌酶对hHSV-1的抑制效果。结果:HEWL对细胞的最大无毒浓度(TC0)和半数中毒浓度(TC50)均>1 000μg·mL-1,HEWL对hHSV-1抑制作用的最小有效浓度(MIC)为104.17μg·mL-1,半数有效浓度(IC50)为46.88μg·mL-1,治疗指数(TI)为10.67。在平行对照试验中,更昔洛韦对BHK-21细胞的TC0和TC50均>500μg·mL-1。更昔洛韦对hHSV-1抑制作用的MIC为7.81μg·mL-1,IC50为3.91μg·mL-1,TI为64。0.5%SDS完全阻断了HEWL的抗病毒能力。结论:蛋清溶菌酶对HSV-1有较明显的抑制作用,作用强度与溶菌酶的浓度相关。

     

    Abstract: Objective: To observe the antiviral effect of hen egg-white lysozyme(HEWL) against herpes simplex virus type 1(hHSV-1).Methods: The inhibitory activity of HEWL against HSV-1 was tested in vitro on BHK-21 cells,using Genciclovir as positive control and 0.5% SDS as the block-reagent.Data elaboration was carried out by calculation of the cytopathic effect(CPE).Results: Both the maximum nontoxic concentration(TC050) of HEWL on BHK-21 cells were more than 1 000 μg·mL-1,the mininum inhibitory concentration(MIC) of HEWL to hHSV-1 was 104.17 μg·mL-1,50% inhibitory concentration(IC50) was 46.88 μg·mL-1,therapeutic index(TI) was 10.67.In the parallel contrast tests,both the TC050-1.the MIC of Genciclovir to hHSV-1 was 7.81 μg·mL-1,IC50 was 46.88 μg·mL-1,TI was 64.0.5% SDS blocked the antiviral activity of HEWL completely.Conclusion: HEWL inhibits the activity of hHSV-1 and the inhibitive effect is proportional to enzyme concentration.
