烫伤大鼠组织白介素-1β转换酶mRNA的表达及其对IL-18和Fas配体的影响

The dynamic change of ICE mRNA expression and its effect on IL-18 and FasL in thermal injured rats

  • 摘要: 目的: 探讨烫伤后肠、肺组织白介素1β转换酶(IL 1βconvertingenzyme,ICE) mRNA的表达对IL 18、Fas配体mRNA表达的影响。方法: 采用大鼠30%体表面积Ⅲ度烫伤模型,组织ICE、IL 18、FasLmRNA含量采用逆转录聚合酶链式反应检测。结果: 烫伤后组织ICE、IL 18、FasLmRNA表达较伤前值即有显著升高,烫伤后8h达高峰且一直持续至伤后2 4h,给予SDD预治疗后可不同程度抑制ICE、IL 18、FasLmRNA的表达。相关分析显示,组织ICE、IL 18、FasLmRNA表达呈显著正相关关系。结论: 组织ICEmRNA表达在烫伤早期即显著增多,并呈一逐渐升高的趋势,对IL 18、FasLmRNA基因表达具有重要影响。

     

    Abstract: Objective: To investigate the dynamic changes of ICE mRNA expression and the relationship between ICE mRNA and IL 18-FasL in thermal injured rats after delayed resuscitation..Methods: The animals were subjected to a 30%total body surface area full thickness thermal injury,fluid resuscitation were performed at 6 hours post burns 54 male Wistar were randomly divided into three groups: normal controls,30% Ⅲ injury group,and selective decontamination of the digestive tract (SDD)treatment group Tissue ICE- IL 18- FasL mRNA expression was determined by reverse transcription po1ymerase chain reaction (RT PCR)..Results: ICE-IL 18-FasL mRNA expression significantly increased at 2 hours,peaking at 8 hours,and a high level was maintained till 24 hours.However, treatment with SDD could markedly inhibit ICE-IL 18-FasL mRNA expression in various organs.In addition, positive correlation between ICE mRNA and IL 18-FasL mRNA expression were noted(r=0.736~0.864, P<0 01)..Conclusion: This study suggests that major burns can lead to a marked elevation of tissue ICE mRNA expression in vital organs,which might be associated with IL 18-FasL mRNA expression.
