EGFR反义重组腺病毒治疗乳腺癌的实验研究

摘要: 目的: 探讨表皮生长因子受体(EGFR)反义重组腺病毒AdE5对乳腺癌细胞移植瘤的作用。方法: 人乳腺癌细胞MDA-MB-231接种于裸鼠右侧背部皮下形成肿瘤后,以AdE5、空病毒载体AdCO1瘤内注射,每只109pfu/100μl。共注射2次。定期测量肿瘤体积,计算相对生长率。肿瘤组织HE染色后做组织学观察,以免疫组化分析K i-67表达。结果: AdE5瘤内注射可抑制肿瘤生长(相对生长率<1)。组织学研究显示肿瘤内坏死明显,免疫组化分析发现AdE5治疗后肿瘤K i-67指数显著低于AdCO1及PBS组。结论: 以腺病毒载体介导的EGFR反义RNA转导可有效地抑制乳腺癌细胞的在体肿瘤生长。Abstract: Objective: To investigate the anti-tumor effects of a recombinant EGFR antisense adenovirus,AdE5,for breast cancer cells. Methods: Human breast cancer MDA-MB-231 cells were injected s.c.into right flanks nude mice.Two intra-tumoral injections of 100μl PBS buffer containing 109 pfu of AdE5 or AdCO1 or PBS alone were practised.The growing tumors were measured,and relative gowth index were caculated.HE coloration was performed for histological study,and Ki-67 expression was determined by immunohistochemical staining. Results: The tumor growth was inhibited by AdE5 treatment(relative gowth index < 1).Histological analyses revealed that AdE5 caused an increase of central necrosis in tumor tissues.Immunohistochemical study showed that Ki-67 expression was significantly inhibited by AdE5 compared with AdCO1 or PBS. Conclusion: The transduction of EGFR antisense RNA by adenoviral vector is effective for inhibition of in vivo growth of breast cancer cells.
