胰岛素样生长因子-1对血管异位钙化作用的研究

Insulin-like growth factor-1 increased the calcium deposition of vascular calcification in vitro

  • 摘要: 目的: 胰岛素样生长因子(IGF-1)对动脉粥样硬化的形成和发展及骨骼组织的矿化均起重要作用,鉴于动脉钙化广泛存在于动脉粥样硬化病变中而且与骨骼矿化过程的相似性,本研究试图研究IGF-1在动脉钙化中的作用。方法: 体外培养牛主动脉平滑肌细胞,建立血管钙化模型,在培养液中加入不同浓度的IGF-1,观察细胞钙沉积变化,以及与之密切相关的细胞碱性磷酸酶活性和骨钙素分泌情况。结果: 10-9mol/L的IGF-1培养10d后,普通对照培养细胞的钙沉积无明显变化,钙化培养细胞的钙沉积增加了11.82%(P>0.05),在此过程中细胞内碱性磷酸酶活性增加了16 8. 7%,骨钙素分泌增加了5 8.3%;增加IGF-1的浓度至10-8mol/L和10-7mol/L,细胞的钙盐沉积进一步增加,分别提高了22.74%(P<0.05)和40.7%(P<0.01)。结论: IGF-1可呈剂量依赖性的促进钙化血管细胞的钙质沉积,其作用可能与促进钙化血管细胞合成碱性磷酸酶及促进细胞向成骨细胞转化有关。

     

    Abstract: Objective: Accumulated evidence suggested that vascular calcification share many characteristics with the minaralization of bone. IGF-1can enhance the minaralization of osteoblastic cell in bone and is also one of the important growth factors in blood vessel. So it is rational to study its role in the vascular calcification. Methods: β-glycerin-phosphate disodium salt was used to induce cultured bovine vessel smooth muscle cell to calcify which serves as a vascular calcifying model in vitro. 10-9mol/L IGF-1were add to the culture media. After 10days culturing,the deposition of calcium,alkaline phosphatase activity and osteocalcin concentration was quantified. Results: After 10days of culture,10-9mol/L IGF-1increased the calcium deposition of calcified BVSMC by 11. 82%(P>0. 05),this processes is time dependent. The cellular ALP activity was dramatically increased with the highest in the 8day with the greatest increase of 168. 7%. The osteocalcin concentration of cultured media syntheses by BVSMCs also elevated 58. 3%. Change the concentration of IGF-1 to 10-8 mol/L and 10-7 mol/L,the calcium deposition of BVSMCs increased by 22. 74% (P <0. 05) and 40. 7% (P <0. 01) respectively. Conclusion: IGF-1 increased calcified BVSMCs'calcium deposition. This may through the stimulating the cellular synthesis of ALP and osteocalcin which indicate that it promote the calcified BVSMCs'changing to osteoblastic cell.
