应用基质辅助激光解吸质谱的源内衰变及串联质谱技术测定重组人纽兰格林的C端序列

Detection of recombinant human neuregulin C-terminal sequences by in-source-delay of MALDI-TOF-MS and LIFT of TOF-TOF-MS

  • 摘要: 目的 测定重组人纽兰格林的C端序列。方法 用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)的源内裂解(ISD)及串联质谱技术(TOF-TOF,LIFT),测定重组蛋白纽兰格林的C端序列。结果 用该方法鉴定了重组蛋白纽兰格林C端序列的10个氨基酸残基。结论 与现有的C-端序列测定方法比较,该方法简便快捷,可适用于部分重组蛋白药物的质量控制。

     

    Abstract: Objective To detect the recombinant human neuregulin(rhNRGL) C-terminal sequences. Methods Ten amino acid residues of rhNRGL C-terminal sequences were detected by in-source-delay(ISD) of MALDI-TOF-MS and LIFT of TOF-TOF-MS. Results The 10 amino acid residues of rhNRGL C-terminal sequences were detected. Conclusion This method is simpler and rapider than the commonly used method and can be applied to quality control of some recombinant proteins.
