低密度脂蛋白受体基因多态性对微粒化非诺贝特调脂疗效影响的研究

Effect of the gene polymorphism of low density lipoprotein receptor on the efficacies of the lipids regulation of micronized fenobrite

  • 摘要: 目的:探讨低密度脂蛋白受体(LDLR)基因HincⅡ酶切位点多态性对微粒化非诺贝特调脂疗效的影响。方法:血清总胆固醇(TC)≥5.20mmol/L和/或甘油三酯(TG)2.00~5.65mmol/L之间者,共52例,随机分为药物组30例和安慰剂组22例。结果:根据LDLR基因HincⅡ酶切多态性分层比较微粒化非诺贝特和安慰剂的调脂疗效,发现药物组和安慰剂组三种基因型(H1H1、H1H2和H2H2)患者之间的TC、LDTC和TG下降的有效率(TC和LDLC下降分别≥10%,TG下降≥20%)无明显差异(P均>0.05);不同基因型之间的TC和LDLC下降幅度亦无显著差异(TC依次平均下降30.3、30.2和33.4%,LDLC依次平均下降28.0、29.1和31.8%,P均>0.05);对血清TG的观察结果亦是如此。说明具有不同LDLR等位基因不论对微粒化非诺贝特的降低血清TG作用,还是对降低血清胆固醇作用均无明显影响。结论:LDLRHinc Ⅱ酶切位点不参与微粒化非诺贝特调整血清胆固醇和TG的作用。

     

    Abstract: Objective: To observe effect of the gene polymorphism of low density lipoprotein receptor on the efficacies of the lipids regulation of micronized fenobrite.Methods: Fifty two patients with hyperlipidemia (serum total cholesterol level exceeded or equal to 5.20 mmol/L, and/or triglyceride level was in the range 2.00-5.65 mmol/L) were randomly allocated into Fenobrite group and paacebo group (30 and 22 cases, respectively). The low density lipoprotein receptor HincⅡ polymorphism were measured by polymerase chain reaction technology.Results: Comparing the efficacies of regulation of lipids of Fenobrite and placebo among the subgroups with H1H1, H1H2, or H2H2 genotype of low density lipoprotein receptor HincⅡ polymorphism, we did not find that the efficacy rates (10% fall in total cholesterol and in low density lipoprotein cholesterol, 20% fall in triglyceride, and increase more than 0.1 mmol/L in high density lipoprotein cholesterol, respectively) of patients with H1H2 or H2H2 genotypes were significantly different among the different genotytes of Fenobrite and control groups. The lowered ranges of total cholesterol, low density lipoprotein cholesterol, and triglyceride were neither marked different among the patients with H1H1, H1H2, and H2H2 genotypes, average reduction percent of total cholesterol were in sequence 30.3, 30.2, and 33.4% (P>0.05), low density lipoprotein cholesterol were 28.0, 29.1, and 31.8% (P>0.05), triglyceride were 55.2, 53.9, and 53.62% (P>0.05). Responsiveness were less different between patients with h1 allele and h2 allele in Fenobrite and placebo group. Conclusion: The result of present study suggested that low density lipoprotein receptor HincⅡ alleles do not took part in the key processes of the cholesterol or triglyceride regulation of Fenobrite.
