TGFβ1基因直接瘤内注射后的表达及其对小鼠肿瘤的影响

TGF β1 gene expression and its effect on murine tumor following direct intratumoral administration

  • 摘要: 用肺腺癌细胞株LM3小鼠背部皮下接种成瘤。于瘤体内多次多点直接注射pMAMneo-TGFβ1质粒DNA,并设空质粒和盐水注射对照组进行比较。发现TGFβ1基因直接注射组肿瘤生长速度增快,肿瘤组织内结缔组织间质明显增多,但转移发生频率的差异无显著性意义。取新鲜的肿瘤组织提取RNA,分子杂交证实,直接注入肿瘤组织内的TGFβ1基因获得了高效表达。结果提示,DNA直接注射法作为invivo基因转移手段有其不可低估的应用前景。

     

    Abstract: Plasmid DNA containing transforming growth factor β1 (TGFβ1) gene was directly injected into subcutaneous nodules of murine lung adenocarcinoma in T 739 mice. Post injection observation showed that the growth of tumor was significantly boosted in the treated mice as compared with the control groups (pMAMneo DNA or saline), whereas there was no significant difference in metastatic behavior. Both Northern and slot blot confirmed efficient expression of TGFβ1 mRNA in the treated group but not in the control. The results suggest that direct intratumoral injection of nude plasmid DNA might be a promising gene transfer strategy in vivo.
