APA微囊免疫隔离效应的体外测定

Determination of immuisolation effect of APA microcapsules in vitro

  • 摘要: 目的:在体外了解海藻酸钙-多聚赖氨酸-海藻酸钙微胶囊(APA)的免疫隔离效应。方法:用下列方法测定APA微囊的免疫隔离效应,用激光共聚焦显微镜测定荧光素标记的IgG微囊膜的扩散;NK细胞对裸露或微囊化K562细胞的杀伤作用;BCCs刺激小鼠脾淋巴细胞增殖的保护效应;IL-2及TNF-α对共培养的细胞或微囊化细胞的杀伤作用。结果:微囊具有阻断IgG免疫球蛋白扩散作用;并且有效地保护囊内K562细胞不受NK细胞的杀伤;防止BCCs刺激小鼠脾淋巴细胞增殖反应;IL-2TNF-α均可通过APA微囊膜,支持IL-2依赖细胞CTLL-2的生存;及对囊内的L929靶细胞产生杀伤作用。结论:APA微囊膜可有效地隔离抗体及淋巴细胞介导的体液和细胞免疫反应。

     

    Abstract: Objective: to understand immuisolation effect of Alginate polylysine alginate(APA) in vitro. Materials: Checking the diffusion through APA microcapsule membrane of IgG labeled with fluorescein by laser scan confocal microscope(LSCM). Assessing injury effect of NK cells to K562 or encapsulated K562. Assessing protection effect of APA microcapsule from mouse spleen lymphocyte proliferation stimulated by BCCs. Checking injury effect of IL-2 and TNF α to cells or microencapsulated cells. Result s: IgG can not go through into APA microcapsule.NK cells have cytotoxic effect to K562 cells and APA can protect K562 from attack of NK. APA can prevent mouse spleen lymphocyte proliferation stimulated by BCCs.IL-2 can pass through APA membrane and support existence of CTLL cells. TNF αcan pass through APA membrane and kill L929 cells. Conclusion: APA microcapsule can remarkablely prevent cell-mediated and antigen mediated immunologic reactions.
