Wistar大鼠C6胶质瘤动物模型的肿瘤生长特点

Growth character of model of Wistar rat cerebral glioma established by inoculation of C6 cells

  • 摘要: 目的: 探讨Wistar大鼠颅内及皮下接种C6胶质瘤细胞的肿瘤生长特点及病理特征。方法: 第一组Wistar大鼠及SD大鼠各5 0只,应用立体定向方法将10 6的C6细胞接种于大鼠脑尾状核,对比观察接种后不同大鼠生存时间及病理特征。第二组140只Wistar大鼠于颅内接种后3d、5d、7d、9d、11d、13d和15d各处死2 0只,观察肿瘤大小及病理学变化。第三组5 0只Wistar大鼠皮下接种2× 10 6的C6细胞,观察皮下肿瘤生长特点及病理特征。结果: Wistar大鼠颅内接种后平均生存期为(15 9± 1 2) d,瘤组织向正常脑组织浸润生长,无明显分界,有肿瘤血管,坏死,卒中发生,S 10 0及GFAP染色阳性。SD大鼠平均生存期为(18 9± 1 8) d,瘤组织与正常脑组织有明显分界。Wistar大鼠颅内接种后第5~11d肿瘤生长最为迅速。Wistar大鼠皮下接种后肿瘤生长稳定,但接种后2 5d停止生长并逐渐缩小。结论: Wistar大鼠颅内C6胶质瘤模型特征更加接近于人恶性脑胶质瘤,Wistar大鼠皮下接种C6细胞肿...。

     

    Abstract: Objective: C 6 intracerebral and subcutaneous model of Wistar rat was established to study the tumor growing characters and the histological characters.Methods: First group, 10 6 C 6 cells were inoculated into caudate nucleus of 50 Wistar rats and 50 SD rats by stereotaxic method to study the survival time and histological characters respectively Second group,140 Wistar rats were killed in batchs 3,5,7,9,11,13,15 days after intracerebral inoculation,the tumor size,the histological characters were observed Third group, subcutaneous model of 50 Wistar rats was established by inoculation of 2 10 6 C 6 cells,the tumor size,the histological characters were observed.Results: The mean survival time after intracerebral inoculation of Wistar rats and SD rats was (15 9±1 2) days and (18 9±1 8) days respectively,the intracerebral tumor of Wistar ratshow several characteristic features of spontaneus glioblastomas,such as neovasculature,parenchymal invasion,nuclear pleiomorphism and presence of hemorragic and necrotic areas,GFAP and S 100 protein immunohischemistry were positive, the intracerebral tumor of SD rats show there was border between the tumor and cerebral tissue The intracerebral tumor of Wistar rats grew faster during 5 11 day after inoculation The subcutaneous tumor of Wistar rats grew steadily until 25 days after inoculation,then the tumor cease growing and the tumor size decreased Conclusions: C 6 intracerebral model of Wistar rats was more similar to human glioma growing characters, C 6 subcutaneous model of Wistar rat had character of selflimitation
