Id蛋白N末端突变体真核表达载体的构建及其三维空间模型预测

Construction of Id mutant proteins in pcDNA3.1(+) vectors and three-dimensional protein structures prediction

  • 摘要: 目的:构建Id家族蛋白N末端突变体的真核表达载体,并模拟其突变前后的三维空间模型。方法:用带有突变的引物从质粒中扩增出两段突变序列,用拼接PCR的方法将两段突变序列合成一条N末端包含两个突变点的Id序列。应用Swiss-PdbViewer3.7(SP5)软件模拟其突变前后的三维结构模型。结果:构建了一系列Id家族N末端突变体的pcDNA3.1(+)真核表达载体。预测出其三维结构的变化。结论:构建Id蛋白家族N末端突变的pcDNA3.1(+)真核表达载体,为进一步研究Id蛋白N末端结构的生物学功效奠定基础。

     

    Abstract: Objective: To construct N-terminal mutants of Id proteins into pcDNA3.1(+) vectors and propose the available three-dimensional protein structures.Methods: Amplifications were carried out with mutagenic primers,and then DNA products were ligated to achieve two points mutant sequences.Three-dimensional protein structures were obtained by Swiss-PdbViewer3.7(SP5) to compare the changes of protein structures.Results: The mutants of Id proteins were cloned into pcDNA3.1(+) vectors successfully,and the changes of their protein structures were predicted.Conclusion: N-terminal Mutants of Id proteins in pcDNA3.1(+) vectors are convenient tools for in-depth studies on the function of Id protein family.
