持续性高正加速度重复作用导致大鼠损伤的自然修复模型

Establishing an animal model for the post-injury regenerating ability of rats exposed to the repeated +Gz forces

  • 摘要: 目的: 建立持续性高正加速度(+G z)重复作用导致大鼠损伤的自然修复模型。方法: 雄性W istar大鼠108只随机分成9组,每组12只。A组为空白对照;B组地面固定5 m in;C组固定于离心机转臂上,俯卧位,头向轴心,+1G z离心5 m in;D组、E组、F组、G组、H组、I组体位同C组,+10G z离心30 s,间隔时+1G z离心60 s,连续5/d,每周4d,共4周。A、B、C、D组于离心次日处死,其余各组分别于离心后1周、2周、4周、8周和12周后次日处死,取颞下颌关节、咀嚼肌、心、肝、肾等器官组织作光镜、电镜观察。结果: ①动物全部成活。②A组、B组和C组器官组织未见异常;其余各组除肾脏未见明显异常外,心脏、肝脏和颞下颌关节、咀嚼肌呈现不同程度病理改变,随时间推移,上述损伤逐步减轻、呈现修复改变。结论: 本动物模型重复性好,实验证实+G z可以导致心脏、肝脏和颞下颌关节、咀嚼肌损伤,并能自然修复,是研究+G z导致机体损伤和自然修复较理想的动物模型。

     

    Abstract: Objective: To analysis the Post-injury Regenerating Ability of rats exposed to the repeated +Gz forces,A animal model was established.Methods: A hundred and eight male rats were randomly assigned to 9 groups with 12 rats in each group.Group A was normally fed as the blank control group.Group B was only fixed with rat-kept devices for 5 minutes as the fixed control group.Group C was borne +1Gz for 5 minutes.In the same position as group C,group D,E,F,G,H and I were repeatedly exposed to +10Gz for 30 seconds each time with 5 times a day and +1Gz for 1 minutes as a interval;the 6 groups were treated in the condition above for 4 days a week and 4 weeks in total.The rats in group A,B,C and D were killed on the next day after the centrifuge.The others in group E,F,G,H and I were killed at the week 1, 2,4,8 and 12 after the centrifuge,respectively.After the rats were killed,temporomandibular joint,masticatory muscles,heart,liver and kidney were taken for(observe) under optical microscope,scanning electron-microscope(SEM) and transmission electron-microscope(TEM).(Results: All) rats survived before killed.There was no significant difference observed in every organ and tissue in group A,B and C.Heart,liver,temporomandibular joint and masticatory muscles showed pathological changes to different extent in the other groups,except kidney.As time going,the above injury weakened and regenerated gradually.Conclusion: The animal model,which has good repetition,is a perfect model to study the Post-injury Regenerating Ability of rats exposed to the repeated +Gz forces.It was confirmed that the repeated +Gz could induce the injury of temporomandibular joint,masticatory muscles,heart and liver,which could be self-regenerated in different period after.
