Abstract:
Objective To study the polypeptide 7-induced immune response of dendritic cells(DC) in mice and its mechanism.
Methods DC were cultured with the "bulk" culture method.Splenocytes were collected from the mice 10 days after subcutaneous immunization with polypeptide 7.CD4
+ T cells were isolated from splenocytes by cell sorting and incubated for 24 hours.Cytokines in supernatants were detected by flow cytometry.
Results The expression rate of CD11c on surface of DC was over 80%.The CD11c showed a typical morphology of DC.The level of TNF-α was lower while the levels of IL-5,IL-2 and IFN-r were higher in supernatant of CD4
+ T cells from experiment group than from control group(
P<0.05).
Conclusion A large number of immature DC can be cultured in vitro with the "bulk" culture method.Peptide 7 can induce immune response of Th1 cells.
