脑红蛋白基因在大脑和不同组织细胞中的表达

  • 摘要: 目的: 检测脑红蛋白(NGB)基因在大鼠脑组织和人体不同组织细胞中的表达。方法: ①wistar大鼠,取脑固定于4%多聚甲醛,提取纯化质粒、NGBcRNA探针标记,原位杂交组化法检测NGBmRNA在正常大鼠脑中的分布。②人的13种组织总RNA各取2ug,逆转录成cDNA,1ul作为模板做PCR。设计间并性引物,逆转录成cDN产物进行琼脂糖凝胶电泳后,定量分析电泳结果。单因素方差分析处理数据。结果: ①NGBmRNA阳性细胞主要定位于神经元、扣带、梨状皮质、海马各区、丘脑、小脑等。②人13种组织中有NGBmRNA的表达,不同组织NGBmRNA扫描的相对光密度值(OD)不同,胎肾、肝、大脑皮层高于全脑,提示除脑组织外机体其他组织亦有NGBmRNA的表达。结论: NGB基因在脑内有广泛的分布,其蛋白质及mRNA主要定位于神经元的细胞质,不同脑区的NGBmRNA阳性细胞的密度不同。人体不同细胞组织中也有NGBmRNA的表达,提示NGB基因并不是神经系统所特有的,NGBmRNA可能在机体较为广泛的区域中发挥着作用。

     

    Abstract: Objective: To study expression of neuroglobin (NGB) gene in brain and different cellular tissues. Methods: 1.Wistar rats, the brian was taken and fixed in 4% polymerisatum at once. plasmid was purificated and extract of NGB, distribution of NGBmRNA was detected in normal rat brain with hybridization in situ and histochemistry method. 2.Total species tissues RNA 2ug of eath tissue,it was been reverse transcription cDNA and PCR templat. infraequally primer was designed, cDNA was been reverse transcription, PCR products was used agarose gel electrophoresis and quantu analysed with software. mono agent analysis of variance of Stat statistical. Results: 1.Positive cell of NGBmRNA localized neuron in normal rat brain、cinguli cortex、pyriform cortex、cytoplasm、hippocampal area、prosencephalon nucleus mass、almond central nucleus、almond inside nucleus、almond cortex pronucleus、almond cortex postnucleus、piriform intranucleus ether、thalamus. 2.There were expression of NGBmRNA in human 13 species besides brain tissue. expression were difference in those tissues. Conclusion: Wide distribution of NGB gene in rat brain, Positive cell of NGBmRNA localized neuron in normal rat brain; There were expression of NGBmRNA in human 13 species cellular tissues, but expression were difference levels in those tissues. It was suggested that NGBmRNA may be to have comprehensive action in human body besides in nervous system play an important rule.
