Role of microRNA-25 in invasion and metastasis of laryngeal squamous cell carcinoma cells

  • 摘要:   背景  导致喉癌患者死亡的主要原因是复发和转移,喉癌侵袭转移的分子机制已成为当前研究的重点。  目的  研究microRNA-25(miR-25)在喉鳞癌Hep-2细胞体外侵袭转移中的调控作用。  方法  应用微小RNA过表达载体(mimic)和抑制载体(inhibitor)转染Hep-2细胞,分别增加和抑制Hep-2细胞中miR-25的表达,Transwell法和细胞划痕实验检测Hep-2细胞侵袭迁移能力的变化,MTT法检测细胞增殖能力变化,绘制细胞生长曲线。   结果  上调Hep-2细胞中miR-25表达后,Transwell实验和细胞划痕实验显示Hep-2细胞的体外侵袭和迁移能力显著低于正常对照组(P<0.01),MTT检测结果显示过表达miR-25组Hep-2细胞增殖能力亦显著降低。而抑制miR-25表达后,Hep-2细胞体外侵袭、迁移和增殖能力均显著增强。  结论  miR-25的表达量显著影响喉鳞癌Hep-2细胞体外侵袭、迁移和增殖能力,提示miR-25可能是抑制喉鳞癌转移的一个有效靶点。
