The effects of angiotensin Ⅱ on the viability of cardiac myocyte

  • 摘要: 目的: 旨在观察血管紧张素Ⅱ(ANGⅡ)对心肌细胞活力的影响。方法: 本研究利用培养的乳鼠心肌细胞,根据活细胞线粒体在能量代谢过程中可作用于MTT产生蓝紫色结晶产物formazan这一原理,应用MTT法测定ANGⅡ对培养心肌细活力的影响。结果: 发现ANGⅡ在12h之内使formazan产物的OD值逐渐上升,12h达到高峰,此后开始下降,至2 4h已低于正常水平,它们的OD值之间均有显著的统计学差异(P<0.0 5或0.0 1)。结论: 该结果提示ANGⅡ在早期具有提高心肌细胞活力的效应。这一发现对了解ANGⅡ在心功能不全代偿和心衰发生中的作用地位具有重要意义。

     

    Abstract: Objective: Our study is to investigate the effects of Angiotensin Ⅱ (ANGⅡ) on the myocyte viability and the cellular basis of myocardiocyte dysfunction, in order to provide the evidences for better management of heart failure.Methods: Based on that the mitochondria of viable cells can act on MTT to produce formazan with light purple, we investigate the effect of ANGⅡ on the cultured myocyte viability by means of colorimetric method.Results: Our results showed that ANGⅡ caused the gradual increase of formazan optical density (OD) in 12 hours, and the formazan OD reached the peak at 12 hour, thereafter decreased, even lower than the normal at 24 hour. CV11974 did successfully and significantly antagornized the effect of AngⅡ on the cardiac myocyte viability (P <0 05 or 0. 01). Conclusion: Our results indicated that AngⅡ could improve the myocytes viability at the early stage. It has important meanings for the correct use of ANGⅡ and the efficient management of heart failure in the future.
