低氧促进大鼠骨骼肌成肌细胞体外增殖

Hypoxia enhances the proliferation of rat skeletal myoblast in vitro

  • 摘要: 目的: 探讨轻中度低氧对大鼠骨骼肌成肌细胞(SkM s)增殖的影响。方法: 采用血球计数板计数法和流式细胞术观察了3%、10%氧浓度及CoC l2对SkM s数量和增殖指数的影响。结果: 低氧组细胞较对照组细胞数量和增殖指数明显增加(P<0.05),10%氧浓度促增殖作用更为明显,CoC l2对大鼠SkM s没有促增殖作用。结论: 轻中度低氧可以促进大鼠SkM s的增殖,为体外大量扩增细胞提供了新思路。

     

    Abstract: Objective: To explore the role of hypoxia in rat skeletal myoblastic proliferation.Methods: We observed theeffects of hypoxia(10%O2、3%O2) and CoCl2on the proliferation of rat skeletal myoblasts.Cell numbers were determined byhemacytometer count.The proliferation indexes were detected by flowcytometer.Results: The numbers and proliferation indexeswere higer(P<0.05) in physiological hypoxia than in those of control group.10%O2hypoxia had more obvious effect.CoCl2had no effect on proliferation of these cells.Conclusion: Hypoxia can promote the proliferation of rat skeletal myoblasts.It is pos-sible to provide a new way for abundantly cultivating these cells.
