自发性高血压大鼠心肌组织蛋白质谱差异分析

Myocardium in rats with spontaneous hypertension:An analysis of proteomic profile

  • 摘要: 目的 研究自发性高血压大鼠蛋白质表达谱的改变。方法 取自发性高血压大鼠和正常血压对照WKY大鼠左心室游离壁心肌组织,提取全部蛋白质后以双向电泳分离蛋白质,经考马斯亮蓝染色、图像分析后选取7个差异在2倍以上的蛋白质点进行肽质量指纹图谱分析。结果 双向电泳可分离454±45个蛋白质,点平均匹配率为91.1%。其中27种蛋白质表达差异2倍以上,质谱鉴定出的6种蛋白质分别为与能量代谢相关的丙酮酸脱氢酶E1α和肌酸激酶,与细胞应激相关的葡萄糖调节蛋白58和αB-晶体蛋白,与细胞生长、凋亡相关的三结构域蛋白50(Tripartite Motif Protein 50,TRIM 50)和LIM-only蛋白家族成员LIM domains(FHL)。其中除肌酸激酶表达下调外,其他5种均较正常血压WKY大鼠上调。结论 自发性高血压大鼠在出现明显的血压改变和心肌肥大之前已经出现能量代谢、应激和细胞生长调控相关蛋白质表达的改变。

     

    Abstract: Objective To study the changes in proteomic profile of myocardium in rats with spontaneous hypertension(SH).Methods Whole tissue protein was extracted from free wall of left ventriclar myocardium of rats with SH and normal control Wistar Kyoto(WKY) rats,and separated by 2-dimensional electrophoresis(2-DE).After stained with Coomassie brilliant blue,7 differentially expressed protein spots were identified by flight mass spectrometry(MALDI-TOF) and then subjected to mass fingerprint analysis.Results 2-DE could separate 454±45 proteins with an average spot matching rate of 91.1%.Twenty-seven proteins were differentially expressed in myocardial tissue from rats with SH.The 6 proteins identified by MALDI-TOF were energy metabolism associated pyruvate dehydrogenase E1α and creatine kinase,cell stress associated glucose-regulated protein 50,alpha B-crystallin,cell growth associated tripartite motif protein 50 and 4,and a half LIM domain.Of these 6 proteins,the creatine kinase level was lower while the levels of the other 5 proteins were higher in rats with SH than in normal control rats.Conclusion Energy metabolism,stress and cell growth-related protein expression are significantly changed in rats with SH before the presence of increased blood pressure and myocardial hypertrophy.
