全反式维甲酸对恶性黑素瘤A375细胞VEGF表达的影响

Effect of all-trans retinoic acid on VEGF expression in human malignant melanoma A375 cells

  • 摘要: 目的 研究全反式维甲酸(ATRA)对体外培养恶性黑素瘤A375细胞VEGF表达影响。方法 应用Real-time PCR检测A375细胞VEGF-B在基因水平的表达。应用ELISA法检测A375细胞上清中VEGF蛋白水平的表达。结果 ATRA可降低A375细胞培养上清的VEGF的mRNA表达和血清浓度,与对照组相比有统计学意义(P<0.05)。结论 ATRA可在mRNA和蛋白水平下调A375细胞VEGF的表达,且这种抑制作用呈时间剂量依赖关系。

     

    Abstract: Objective To investigate the effect of all-trans retinoic acid(ATRA)on level of mRNA and protein expression levels in malignant melanoma A375 cells in vitro. Methods VEGF-B mRNA and VEGF protein expression levels in A375 cells were measured by real-time PCR and ELISA,respectively. Results ATRA decreased the VEGF-B mRNA and VEGF protein expression levels in supernatant of cultured A375 cells(P<0.05). Conclusion ATRA decreases the VEGF-B mRNA and VEGF protein expression levels in A375 cells in a dose-dependent manner.
