CD44蛋白表达与大肠癌淋巴结转移相关性研究

The relation between CD 44 expression and lymph node metastasis of colorectal carcinoma

  • 摘要: 应用CD44单抗对58例石蜡包埋组织(38例大肠癌组织、9例癌旁组织和11例淋巴结组织)进行免疫组化染色并进行图像分析定量研究。结果显示:38例大肠癌,全呈CD44阳性反应,9例癌旁粘膜上皮细胞弱阳性,5例有癌转移的淋巴结呈阳性,6例无癌转移的淋巴结阴性。大肠癌比癌旁正常粘膜的染色强度要深,两者差异有极显著意义(P<0.01)。CD44免疫反应强度与淋巴结有无转移有关,有淋巴结转移(DukesC/D期)较无淋巴结转移的(DukesA/B期)大肠癌反应强,两者差异也具有极显著意义(P<0.01)。结果表明:CD44不仅与大肠癌形成有关,而且在大肠癌淋巴结转移中起重要作用,检测CD44的表达有可能作为临床预测大肠癌淋巴结转移趋势的重要指标之一。

     

    Abstract: paraffin embedded colorectal carcinoma and 9 matched normal colon mucosa and 11 lymph nodes were tested by immunohistochemistry for CD44 expression using CD44 monoclonal antibody and immunohistochemical quantitative analysis using image analyser. The results showed that all colorectal carcinomas were positive and 9 nomal colon mucosa were weakly positive. The five metastatic lymph nodes were positive and the six non metastatis lymph nodes were negative. Statistical analysis revealed that the expression intensity of CD44 was stronger in colorectal carcinomas than in normal colon mucosa (P <0.01). The expression of CD44 was more intense in metastatic (Dukes C/D) than in non metastatic (Dukes A/B) colorectal carcinomas (P <0.01). Our results indicate that the abnomal expression of CD44 is not only involved in the development, but also involoved in metastasis of colorectal carcinomas. Therefore, detection of CD44 expression may have significant clinical importance in evaluation of the tendency of lymphatic metastasis.
