低温储存对皮肤组织α辅肌动蛋白和纽蛋白表达的影响

摘要: 目的: 观察α辅肌动蛋白和纽蛋白在不同温度保存皮肤组织中的分布,以探讨其与皮肤冷冻损伤的关系。方法: 用α-actinin、vinculin抗体对5种保存条件下的皮肤组织作免疫组化染色,用图像分析仪测定表皮组织中的含量,并用HE染色观察皮肤的组织结构变化。结果: 与新鲜皮片组相比,-196℃H抗冻液中保存皮肤组织结构保存较好,α-actinin、vinculin含量变化不明显,而4℃、-20℃、-80℃保存皮肤组织结构破坏明显,含量亦明显下降。结论: 皮肤低温保存冷冻损伤与黏附分子α-actinin、vinculin破坏有关。Abstract: Objective: To find out the distribution of α-actinin and vinculin in cryopreserved human skins and their relationship with cryoinjury.Methods: Immunohistochemical staining with antibodies of α-actinin and vinculin and histopathological observation were used in 5 groups of skins. Results: Comparing to the normal human skin, The histological structure and α-actinin、vinculin level of skins in stored at -196 ℃ in H anti-frozen liquid did not change significantly, while those in groups kept in 4 ℃、-20 ℃、-80 ℃ were destroyed.Conclusion: The skin cryoinjury is correlated with cell adhension molecules.
