福定碱对小鼠学习和记忆过程的促进作用

Effects of fordine on learning and memory processes in mice

  • 摘要: 用Y-型迷路箱测定福定碱对小鼠空间辨认学习和记忆过程的影响。福定碱5~100μg/kg于训练前30分钟、训练后立即及训练后24小时腹腔注射,明显减少达标所需的训练次数,增加主动回避反应。其促进作用呈剂量依赖关系。口腔给药也有类似的效果。电脑休克对小鼠学习记忆能力有损害作用,而福定碱40μg/kg有明显的保护小鼠免受损害的作用,使记忆的获得、巩固及再现三个过程基本上恢复到正常动物的水平。

     

    Abstract: Fordine, a new reversible acetylcholinesterase inhibitor, has been shown in our lab to improve learnig and memory and has therapeutic effect on experimental autoimmune myasthenia gravis and anti-arrhythmic effect induced by epinephrine in animals. By using Y-maze, the effects of fordine on learning and memory processes in discrimination task were studied. Fordine 5-100 μg/kg ip beffore training, immediatly after training and 24 h after training caused significant decrease in the mean number of trials and increase in the active avoidance response when these parameters were measured in normal mice. Electro-brain shock (EBS) severely disrupted the animal ability of learning and memory, but administration of fordine (40ug/kg) markedly protected the animals against the behaviaral effects of EBS, without effect on directly stimulated stun. Fordine prevented the EBS-induced impairment of acquisition, consoliation and retrieval processes. The facilitatory effects of fordine were also seen after oral doses of 10-40 ug/kg. It is suggested that fordine may be a useful compound for the improvement of learning and memory deficits.
