供体脾细胞输注增强CTLA4Ig致小鼠皮肤移植耐受的研究

Study on the effect of combination use of donor-specific-spleen-cell-transfusion(DST) and CTLA4Ig on skin allograft survival in mouse

  • 摘要: 目的: 研究联合应用供者特异性脾细胞输注(donor specific spleen cell transfusion,DST)与CTLA4Ig对促进小鼠皮肤移植物存活的作用。探索CTLA4Ig促进移植物存活的作用机制。方法: 单独或联合应用CTLA4Ig及供者特异性脾细胞输注,同时行皮肤移植,观察皮肤移植物存活时间并检测受者脾细胞对供者细胞的单向混合淋巴细胞反应。结果: 联合应用供者特异性脾细胞输注及CTLA4Ig使小鼠皮肤移植物的存活时间延长,并使受者脾细胞对供者细胞的反应性特异性降低,与未处理组、单用CTLA4Ig或供者特异性脾细胞输注处理组相比有显著性差异(P<0.01)。结论: 联合应用供者特异性脾细胞输注及CTLA4Ig可促进皮肤移植物的存活。移植物中过客淋巴细胞的存在可能对CTLA4Ig诱导免疫耐受有促进作用。

     

    Abstract: Objective: To investigate the effects of combination use of DST and CTLA4Ig to the prolongation of skin allograft survival in mouse and to study the mechanism of CTLA4Ig to prolong the survival of allograft. Methods: Skin allograft recipient mice were treated with CTLA4Ig or DST alone or in combination.Survival time of skin allografts was observed and mixed lymphocyte reaction (MLR) was examined to detect the reactivity of spleen cells of recipient mice to cells from donor mice or third party. Results: Survival time of skin allografts was prolonged in group treated with both DST and CTLA4Ig with the reactivity of spleen cells of recipient mice to cells of donor mice degraded specifically while groups treated with DST or CTLA4Ig alone show no difference to negative control group. Conclusions: Combination use of DST and CTLA4Ig can prolong the survival time of skin allograft in mice.The presence of passenger leukocyte in organ allograft may be helpful to the allograft tolerance inducing effect of CTLA4Ig.
