利用MALDI-TOF MS进行卵巢癌血清诊断模型的建立

Establishing ovarian cancer serum diagnosis model with MALDI-TOF MS

  • 摘要: 目的 利用基质辅助激光解析飞行时间质谱技术(MALDI-TOF MS)进行卵巢癌血清诊断模型的建立及潜在肿瘤标志物的筛选。方法 卵巢癌患者血清20例;健康对照人群血清20例。利用弱阳离子交换磁珠(MB-WCX)进行样品前处理工作,质谱分析应用线性模式,采集血清多肽指纹图谱,建立相关的诊断模型并筛选卵巢癌特异性蛋白质峰。结果 建立了用于进行卵巢癌血清诊断的模型,利用GA、SNN和QC方法分析,认为其灵敏度和特异性都可以达到90%以上,并找到一些差异表达的蛋白质,可以用于进行疾病与正常的区分。结论 MALDI-TOF MS技术是一种能够快速、简便且无创的分析方法,不仅能够用以建立血清诊断模型,还可以用以进行筛选卵巢癌患者血清中相对特异的潜在肿瘤标志物,具有较好的临床应用前景。

     

    Abstract: Objective To establish ovarian cancer serum diagnosis models and to find potential biomarkers with MALDI-TOF MS. Methods Twenty ovarian cancer patients and 20 healthy controls were enrolled to pre-extract the serum samples using the MB-WCX kit.Then,linear mode was used to obtain MALDI-TOF protein profiles,to establish serum diagnosis models,and to find some prominent proteins. Results Ovarian cancer serum diagnosis models were established and some prominent peaks that could be used as potential ovarian cancer biomarkers were found.The sensitivity and specificity were both more than 90%according to GA,SNN and QC methods.Some of the proteins could be used to differentiate diseased people from healthy ones. Conclusion s MALDI-TOF MS is a high-throughput sample preparation method and a potential tool for the diagnosis of human diseases.It can be used not only to establish serum diagnosis models,but also to search for potential biomarkers in ovarian cancer patients′serum samples.Thus it has a good future for routine clinical work.
