培养鼠心肌细胞在移植大鼠体内生长发育的初步观察
Study of fetal cardiomyocytes transplanted into adult myocardium

摘要: 目的: 观察移植心肌细胞在成年大鼠心肌组织内生长发育过程中的形态学变化及其与宿主心肌相互作用情况。方法: 将培养的F34 4近交系乳鼠的心肌细胞,经胶体金标记后,直接注入F34 4大鼠的心肌组织,8周内分别分批取标本,进行组织学和电镜观察。结果: 与移植10d后的结果相比,4周后移植细胞明显体积增大,成熟度增加,并在移植细胞与宿主细胞间见润盘,缝隙连接及桥粒。在8周后的心肌组织切片中,移植心肌细胞的排列方向发生了变化,细胞排列与宿主心肌纤维的方向平行。结论: 移植的心肌细胞在受体心肌内可以继续生长发育,并逐渐分化成熟;移植细胞与受体心肌细胞间可以形成功能性合胞体并能产生与宿主心肌纤维方向一致的收缩活动。Abstract: Objective: To study whether the grafted fetal cardiomyocytes could survive,proliferate and connect with the adult host myocardium. Methods: Cardiomyocytes from F344 fetal rat hearts were grafted into the hearts of adult syngenic recipient mice.The hearts were harvested and processed for histologic and electron microscopic examinations 10 days to 8 weeks after the cell grafts. Results: At 4 weeks,the grafted cardiomyocytes possessed abundunt mature mitochondria and well\|organized myofibrils compared with those at 10 days.Intercalated disks between graft and host cardiomyocytes was observed.The connection consisted of fascia adherents and gap junctions. At 8 weeks,the grafted cardiomyocytes oriented to align themselves parallel to the direction of host myocardium. Conclusion: The grafted fetal cardiomyocytes could differentiate into mature cardiomyocytes and form functional syncytia with the host myocardium in the host heart after cell implantation and the implanted cells might contract synchronously to allow effective cooperation with the host myocardium.
