DCCmRNA表达与大肠癌临床病理特征及PCNA的相关性研究

Corelation analysis of DCC gene mRNA expression in colorectal carcinoma and their clinical pathological characteristics and PCNA

  • 摘要: 应用RT-PCR和免疫组化检测大肠粘膜、腺瘤及腺癌组织DCCmRNA和PCNA表达状况。结果表明:(1)大肠粘膜中均能检出DCCmRNA表达,9例腺瘤1例DCCmRNA表达缺失(11.1%),33例腺癌20例DCCm-RNA表达缺失(60.6%)。(2)大肠癌中DCCmRNA表达缺失与分化程度、浸润深度及转移有显著相关性(P<0.01)。(3)大肠癌DCCmRNA表达缺失与PCNA指数显著相关(P<0.001)。提示:检测大肠癌DCCmRNA表达有助于判断大肠癌的恶性表型。其可能作用机制是DCC基因失活后导致其抑制细胞增殖的特性丧失,从而使大肠癌细胞获得更强的浸润和转移能力。

     

    Abstract: A highly rapid, sensitive and specific method was used for measuring the expression of the DCC gene in clinial samples, based on the polymerase chain reaction. The PCNA expression in colorectal carcinoma tissues was also measured by immunohistochemistry technique. The results showed:(1) There was a normal mRNA expression in 42 colorectal mucosas samples; one out of 9 (11.1%) and 20 out of 33(60.6%) cases lost of DCC gene mRNA expression in adenomas and adenocarcinomas respectively. (2)The lack of DCC gene mRNA expression was closely related with degree of tumor differentiation, depth of invasion of the bowel wall and metastasis(P <0.01). (3)The colorectal carcinomas that lost of DCC mRNA expression have revealed a higher PCNA index (P <0.001). The present study suggests that measuring of DCC mRNA expression is very useful to judge the degree of malignancy, invision and metastasis of colorectal cances. The machinism that DCC gene influences the colorectal carcinoma expressing type may be the loss of its ability in inhibitting cell proliferation when it is inactived.
