逐瘀通脉胶囊对脑缺血大鼠脑组织中IL-1β含量的影响

摘要: 目的: 观察逐瘀通脉胶囊对脑缺血大鼠模型脑组织中白细胞介素-1β(IL-1β)含量的影响,探讨逐瘀通脉胶囊抑制脑缺血损伤级联反应的作用机制。方法: 雄性Wistar大鼠,随机分成假手术对照组、脑缺血组、脑缺血+药物组(逐瘀通脉胶囊组)。采用大脑中动脉栓塞方法,制作脑缺血模型,在光镜下观察各组大鼠脑组织形态学改变。应用免疫组化方法检测脑组织中IL-1β的含量。结果: 脑缺血模型成功后,脑缺血+药物组(逐瘀通脉胶囊组)周围脑组织中IL-1β阳性细胞数,随不同的时间点变化而逐渐增加,但与脑缺血组比较,在不同的时间点均低于脑缺血组,有显著性差异(P<0.05),具有统计学意义。结论: 逐瘀通脉胶囊能够抑制IL-1β的合成与分泌,对脑缺血后的级联反应具有抑制作用。Abstract: Objective: To observe the contribution of the blood circulation capsule to the concentration of IL-β in rat brain tissue with cerebral ischemia and to explore mechanism of the blood circulation capsule which restraint cascade reaction of cerebral ischemia.Methods: The rats were randomly divided into three groups: control group,cerebral ischemia group and drug group.12 and 24 hours models were made through embolisming middle cerebral artery.The morphologic change of rat brain was detected under the light microscope and IL-β in the rat brain tissue was detected by the method of immunohistochemistry.Results: After the model was made,IL-1β positive cells in periphery brain tissue increased gradually at deferent time in cerebral ischemia & drug treated group.But compared with cerebral ischemia group,they were lower.And the diference between the two groups was significantly(P<0.05).IL-β in 12 hours drug group and cerebral ischemia group were higher than control groups(P<0.05).IL-β in 24 hours drug groups was significant lower than cerebral ischemia group(P<0.01).Conclution: Blood circulation capsule can restrant the syhthesis and secretion and has inhibitory effect on the cascade reaction of the cerebral ischemia.
