黄芪总黄酮对BEL-7402细胞的体外抑制作用

Inhibition effects of total flavonoids of astragalus on bel-7402 cell in vitro

  • 摘要: 目的: 探讨黄芪总黄酮(total flavonoids of Astragalus,TFA)在体外实验条件下对人肝癌细胞(BEL-7402)生长、繁殖的影响。方法: 以BEL-7402细胞为实验对象,5-氟尿嘧啶(5-FU)为阳性对照,比较观察了不同浓度的TFA与BEL-7402细胞共同培养不同时间后,TFA对BEL-7402细胞的形态学、细胞存活曲线、细胞生长曲线和细胞有丝分裂指数的影响。结果: 数据表明TFA处理48h半数抑制浓度(IC50)为40μg/m l,5-FU为0.3μg/m l;TFA浓度达80μg/m l时可使BEL-7402细胞生长明显受挫(P<0.05),有丝分裂指数下降;与对照比较细胞形态主要表现为胞浆中颗粒明显增多,胞浆出现大量空泡,核固缩。结论: TFA对BEL-7402细胞生长繁殖具有抑制作用。

     

    Abstract: Objective:To investigate whether the total flavonoids of Astragalus(TFA) has an inhibition property on growth of BEL-7402 cells line.Methods:To study the effect of different concentration of TFA for different culture time with BEL-7402 cell line,the 5-FU as a positive control,the changes(such as morphology,curve of survival,curve of growth and the index of karyokinesis) have been assayed.Results:The results showed that the concentration of the half inhibition after treated 48h was(40μg/ml) and 0.3μg/ml for TFA and 5-FU,respectively.The data also indicated that the TFA have a significant property to inhibit BEL-7402 cell growth(P<0.05) and to reduce the index of karyokinesis when the concentration of TFA was 80μg/ml((P<0.05)).Meanwhile,the morphology BEL-7402 have a significant change such as a lot of vacuole in cytoplasm and ambiguous nucleus after treated by TFA for some time.Conclusion:TFA has a potential effect of inhibition BEL-7402 growth.
