黄芪注射液调节一氧化氮合酶抗再灌注损伤作用

Astragalus injection exerts anti-reperfusion injury through NOS in cultured cardiac myocytes

  • 摘要: 目的:研究黄芪注射液抗培养乳鼠心肌细胞再灌注损伤的作用与一氧化氮合酶(NOS)表达的关系。方法:采用培养的SD乳鼠心肌细胞缺氧复氧模型,实验分为5组:A组为正常对照组,B组、C组、D组、E组均为缺氧-复氧组。各组培养液组成:B组缺氧液,C组缺氧液加黄芪,D组缺氧液加NOS抑制剂L-NAME,E组缺氧液加黄芪加L-NAME。缺氧4 h,复氧2 h。比较缺氧-复氧后培养液中心肌酶含量,并进行心肌细胞NOS蛋白表达检测。结果:复氧2 h后,B组、C组、D组、E组与A组比较心肌酶谷草转氨酶(GOT)、乳酸脱氢酶(LDH)、肌酸激酶(CK)均有升高(P<0.05)。C组与B组比较GOT升高减轻(P<0.05),但无统计学意义。黄芪增强缺氧-复氧引起的心肌细胞NOS表达增加,L-NAME对黄芪引起的NOS表达增加有一定抑制作用。结论:黄芪能够减轻缺血再灌注损伤,其机制涉及了NOS信号转导途径。

     

    Abstract: Objective: To investigate the relationship between the protective effect of Astragalus injection and the expression of nitric-oxide synthase(NOS) in cultured cardiac myocytes.Methods: Studies were conducted on cultured cells.Rat ventricular myocytes were subjected to hypoxia 4 h and reoxygenation 2 h.Astragalus injection was applied to cells during ischaemia/reoxygenation,myocardial enzyme was determined and NOS expression was detected by Western blot after 120 min of reperfusion.Results: In cardiac myocytes,incubation with Astragalus during ischaemia/reoxygenation attenuated the extent of cell damage.Protective effect of Astragalus injection was decreased by L-NAME,an inhibitor of NOS activation,but had no statistical significance.Astragalus enhanced the expression of NOS in ischaemia/reoxygenation myocytes.Conclusion: This study showed that Astraglus attenuated cardiac myocytes injury during reperfusion and the mechanism involved NOS signal pathway.
